Оценка генетических полиморфизмов в популяции Rewa в Центральной Индии с использованием STR-маркеров Y-хромосомы

Цель исследования — проанализировать генотипы мужского населения области Rewa в Центральной Индии с помощью Y-коротких тандемных повторов (short tandem repeats, STR), а затем сравнить результаты с ранее опубликованными данными по другим популяциям.

Материалы и методы. Для исследования был генотипирован 181 мужчина (не родственники) из популяции Rewa по семнадцати локусам Y-STRs (DYS19, DYS389I, DYS389II, DYS390, DYS391, DYS392, DYS393, DYS385a/b, DYS437, DYS438, DYS635648, DYS6458, DYS6458, DYS438, DYS6458, DYS439 и Y-GATA-H4) с использованием мультиплексного набора AmpFlSTR® Yfiler™ (Thermo Fisher Scientific, США). Оценивали частоты аллелей и параметры, имеющие судебно-медицинскую значимость.

Результаты. Идентифицировано 111 различных аллелей Y-STR с частотами в диапазоне от 0,006 до 0,829. Показатели разнообразия генов варьировали от 0,3092 у локуса DYS437 до 0,8188 у локуса DYS385b. Исследуемая популяция показала высокий уровень гаплотипического разнообразия (0,9985) и дискриминационной способности (0,927). Также был проведен гаплотипический анализ. У обследованных мужчин (n=181) обнаружено 165 гаплотипов и 153 уникальных гаплотипа. Кроме того, с использованием анализа молекулярной дисперсии (AMOVA) были рассчитаны значения Rst (genetic distance — генетическое расстояние) для исследуемой популяции и для других 18 популяций, описанных в литературе. Rst представляет собой удобный параметр для оценки уровня генетической дифференциации по микросателлитным данным. На основе этих значений Rst и с использованием графика многомерного масштабирования построено филогенетическое дерево методом соединения соседей.

Заключение. Высокие значения разнообразия гаплотипов и дискриминационной способности указывают на явные генотипические различия мужчин в исследуемой популяции. Можно ожидать, что полученные данные найдут свое применение в судебно-медицинской экспертизе и популяционной генетике.
